干细胞标志物简要介绍

什么是干细胞?

干细胞是一类具有无限的或者永生的自我更新能力 的细胞、能够产生至少一种类型的、高度分化的子代细 胞。大多数生物学家和医学家认为干细胞是来自于胚 胎、胎儿或成人体内具有在一定条件下无限制自我更新与增殖分化能力的一类细胞,能够产生表现型与基因型 和自己完全相同的子细胞,也能产生组成机体组织、器 官的已特化的细胞,同时还能分化为祖细胞。

常见的干细胞标志物 

常见干细胞标志物一般在多种干细胞中表达,常用于鉴别干细胞与其他细胞。Oct4 是干细胞的常见 标志物之一在干细胞维持其多潜能性和自我更新的过程中起着重要作用。Oct4 主要表达于胚胎干细胞 以及生殖干细胞中,它与 Nanog 和 SOX2 通过共同作用,形成一个表达特异性基因的转录网络,从而 维持干细胞的自我更新能力。并目抑制干细胞的过度分化。这些基因在早期发育阶段大量表达,机体成 熟后其表达量随之降低。除此之外 CD30、GATA4、CD133 等也是干细胞的常见标志物,在干细胞各 项生理活动中十分重要。

造血干细胞标志物 

造血干细胞(Hematopoietic stem cells, HSC)是一类成体干细胞,具有干细胞的特性:自我更新 和分化潜能。造血干细胞是一种骨髓来源的多能干细胞,它是血液系统中的“种子”细胞。造血干细胞(HSCs) 在造血过程中形成血系中的所有细胞,包括各种成熟细胞如白细胞、红细胞、血小板等。在受到适当 刺激时还分化为其他非造血组织(如脂肪细胞、心肌细胞、内皮细胞和胰腺细胞)。造血干细胞具有调节体内平衡、免疫功能、抗微生物、抗炎症等生物学功能。它在血液病、遗传性 血液病和自身免疫性疾病的治疗方面有重要作用。CD34是最重要的标志之一,在造血干细胞及其祖细胞的分离和鉴定中,CD34分子是第一个被广泛研究的分子。CD34在人脐带血、骨髓和外周血中的表达约0.1 - 4.9%。CD34在0.55%的人骨髓细 胞上表达,在早期祖细胞上有表达,而在成熟的骨髓细胞上不表达。其他表面标记已经与CD34结合使 用来区分原始细胞群体。此外还有很多常见造血干细胞标志物应用于试验检测中,如CD38、CD90、 CD117、CD123等等。

神经干细胞标志物

神经干细胞(neural stem cell)是指存在于神经系统中,具有分化为神经神经元、星形胶质细胞和 少突胶质细胞的潜能,从而能够产生大量脑细胞组织,并能进行自我更新,并足以提供大量脑组织细 胞的细胞群。是一类具有分裂潜能和自更新能力的母细胞,它可以通过不对等的分裂方式产生神经组 织的各类细胞。需要强调的是,在脑脊髓等所有神经组织中,不同的神经干细胞类型产生的子代细胞 种类不同,分布也不同。常用的神经干细胞标志物有Nestin和SOX2。其他表达于细胞表面的蛋白标志物有ABCG2、 FGFR1和Frizzled-9。Nestin 是一种丝状蛋白,定位干细胞质中,可能在神经元的分化中起一定作 用,在神经干细胞分化初期开始表达,并随着分化程度的加深表达量逐渐降低甚至完全停止。SOX2 主要的作用是维持干细胞增殖和抑制干细胞定向分化,类似于 Nestin,其在成人脑组织中表达量很 低,目前已经作为区分恶性胶质瘤细胞与正常脑细胞的标志蛋白和治疗靶点。此外,该研究领域也发 现了神经干细胞的其他常见标志物,如 CXCR4、Vimentin、NeuroD1、MAP2、Tau 等等。

间充质干细胞标志物

间充质干细胞(mesenchymal stem cells,MSC)是干细胞家族的重要成员,属于成体干细胞。来源于发育早期的中胚层和外胚层。间充质干细胞属混杂细胞群,其表面抗原也具有非专一性,表达 了间质细胞、内皮细胞和表皮细胞的表面标志。目前人类间充质干细胞表面比较肯定存在的标记物有 CD10,CD13,CD29,CD90和CD1,表面阴性的为CD14,CD34,CD45。CD29也被称为整合素β1,VLA-β链或gpIIa,为多种细胞外基质蛋白的受体,是分子量为130 kDa的单链I型糖蛋白,属于整合素家族。它广泛表达于大多数造血细胞和非造血细胞,包括白细胞(虽 然在粒细胞水平较低)、血小板、成纤维细胞、内皮细胞、上皮细胞和肥大细胞。CD29作为纤维连接 蛋白受体,参与多种细胞-细胞和细胞-基质相互作用,调节多种重要的生物学功能,包括胚胎发育、 伤口修复、止血和防止细胞程序性死亡。它的表达与MSC的迁移有关。CD73:CD73是一种5′-核苷酸外切酶,又称NT5E。它是一种69 kDa GPI锚定的表面蛋白。CD73广泛表达于包括淋巴细胞、内皮细胞、平滑肌细胞、上皮细胞和成纤维细胞在内的多种细胞。CD73不仅可参与嘌呤核苷酸的补救合成途径,还可作为一种重要的免疫信号分子,参与跨膜信号转导 及细胞的黏附。CD73在间充质干细胞表面的稳定表达是鉴别MSCs重要的表面标记之一。CD90 (Thy1):CD90也称为Thy-1,是一种25-35kDa 的GPI锚定蛋白。它属于免疫球蛋白超级 家族。人CD90表达于神经细胞、CD34+细胞亚群、胎肝细胞亚群、胎胸腺细胞亚群、成纤维细胞、 活化内皮细胞和某些白血病细胞系。Thy-1与与细胞的黏附、分化、细胞间相互作用有关。它是人类微 血管内皮细胞活化的标记,与新生血管的形成有关;也是鉴别人类MSCs的重要标记之一。
