缺氧信号
动物细胞对氧气的感知和反应是—个非常重要的过程,而缺氧 (Hypoxia) 是指细胞氧气消耗和血管氧气输送失衡,所致的细胞内氧气含量和压力低于正常水平的生理情况。短暂的缺氧可以诱导许多基因的表达,促进组织修复;长期慢性缺氧则会引发功能障碍,造成伤害。在自然界中,生物已经进化出适应缺氧的机制,该过程主要由缺氧诱导因子 (HIF) 复合物进行调控,该复合物在哺乳动物的发育、生理和疾病发病机理中起着至关重要的作用。
缺氧因子
缺氧诱导因子(HIF)是最主要的能够应答细胞内氧气浓度的降低而对多种基因进行调控的转录因子家族,与生物体的生长、发育及一些疾病的发病机理都存在着密切的关系。研究表明,缺氧因子是由α亚基和β亚基组成具有转录活性的异源二聚体。其中α亚基 (HIF-α) 严格受到氧气浓度调控,是缺氧因子的调节亚基,而β亚基 (HIF-β) 则持续性表达。因此,对于缺氧因子的主要研究集中在HIF-α。
细胞在缺氧条件下
在氧气浓度较低时,脯氨酸羟化酶(PHD)没有氧气作为底物,导致HIF-α泛素化无法进行,细胞内的泛素化酶(VHL)无法识别未泛素化的HIF-α进行降解,则导致HIF-α聚集在细胞内。聚集的HIF-α与HIF-β结合,形成有启动转录活性的HIF复合物。在细胞核中,HIF复合物与缺氧反应基因启动子区上的缺氧反应元件(HRE)结合,募集其他转录因子,激发下游的基因的表达。同时,质膜上的多种受体就会通过各种信号通路,上调HIF-α水平,促进缺氧相关基因表达。

细胞在正常氧气条件下
细胞在氧气含量正常的情况下,HIF-α在依赖于氧气的PHD的脯氨酸残基上羟基化和依赖于氧气的HIF1AN的天冬氨酸残基上羟基化。羟基化的HIF-α与VHL肿瘤抑制蛋白(pVHL),将HIF-α进行降解,从而减少HIF-α浓度。此外,在不缺氧气的情况下,HIF-α还会被其他物质抑制转录活性,阻断缺氧信号的传递。
